Миссия состоит в предотвращение террористических актов на территории США, сокращении угрозы терроризма и помощи жертвам терактов. Создано в рамках переоценки стратегии национальной безопасности [ homeland security] в ответ на угрозы международного терроризма, возникшие после событий 11 сентября 2001 [ September 11], в целях координации мер, ранее осуществлявшихся различными ведомствами. Возглавляется министром национальной безопасности [Secretary of the Department of Homeland Security], назначаемым Президентом США "по совету и с согласия" [ advice and consent] Сената [ Senate, U.S.]. Структура включает 4 основных отдела-директората [directorate]: Директорат по безопасности границ и транспорта [Directorate of Border and Transportation Security] (помимо защиты государственной границы и всех видов транспорта, отвечает за исполнение иммиграционного законодательства), Директорат по готовности к чрезвычайным ситуациям и немедленному реагированию [Directorate of Emergency Preparedness and Response] (реагирование на теракты и крупные стихийные бедствия), Директорат по вопросам науки и технологии [Directorate of Science and Technology] (технологическая готовность США к защите от оружия массового поражения в случае его применения террористами), Директорат по информационному анализу и защите инфраструктуры [Directorate of Information Analysis and Infrastructure Protection] (анализ разведывательных и оперативных данных ведомств различных уровней в целях предотвращения угроз терактов). В состав Министерства вошли также Береговая охрана США [ Coast Guard, U.S.], Федеральное агентство по управлению в чрезвычайных ситуациях [ Federal Emergency Management Agency], Секретная служба [ Secret Service, U.S.], Бюро по делам гражданства и иммиграционным услугам [ Bureau of Citizenship and Immigration Services], Администрация по безопасности на транспорте [Transportation Security Administration] (безопасность аэропортов), Таможенная служба США [ Customs Service, U.S.].
